PI3K in Inflammation

The PI3Kδ and PI3Kγ isoforms are strongly implicated in immune-mediated inflammatory and allergic disorders.

  • Unlike other forms of PI3K, expression of the PI3Kδ and PI3Kγ isoforms are restricted primarily to cells of the immune system
  • PI3Kδ and PI3Kγ regulate diverse cellular functions of B cells, T cells, dendritic cells, mast cells and neutrophils
  • Gene targeting and use of prototypic small molecule, selective inhibitors have implicated the PI3Kδ isoform in acquired immunity and the PI3Kγ isoform in innate immunity
  • PI3Kδ-selective, PI3Kγ-selective and PI3Kδ/γ-dual selective inhibitors have demonstrated activity in models of immune-mediated inflammatory and allergic disorders

PI3Kδ/γ-selective inhibitors would be expected to have therapeutic effects in immune-mediated inflammatory and allergic disorders without inhibiting PI3K signaling that is critical to the normal function of healthy cells.
